 Estudió diseño en la Escuela de Artes Industriales del Museo de Filadelfia, de la cual egresó en 1938. Su profesor fue el fotógrafo Alexey Brodovitch, quien más tarde sería su colega en la revista Harper’s Bazaar. Luego viajó a México, donde se dedicó a la pintura durante un año. Sus dibujos fueron publicados en Harper's Bazaar. Su primera labor en la revista Vogue fue como ayudante del artista Alexander Liberman. En 1943, comenzó a trabajar como diseñador de portadas. Después de la Segunda Guerra Mundial, Penn adquirió fama por sus elegantes y glamorosos retratos femeninos publicados en Vogue. En sus fotografías, el sujeto solía posar ante un sencillo fondo blanco o gris, usando la simplicidad más efectivamente que otros fotógrafos de la época. En 1950, se casó con la modelo Lisa Fonssagrives, con quien tuvo un hijo llamado Tom. Tres años después, fundó su estudio fotográfico. Quedó viudo en 1992, cuando Fonssagrives tenía 80 años. Recibió el premio Hasselblad en 1985, y dos años más tarde fue galardonado con el Premio de Cultura de la Asociación Alemana de Fotografía. Publicó diversos libros, incluyendo The astronomers plan a voyage to Earth (1999) y Photographs of Dahomey (2004), además de exhibiciones de su obra. Falleció el 7 de octubre de 2009 en su casa de Manhattan a los 92 años.[1]
